General description
The gene TSP1 (thrombospondin 1) is mapped to human chromosome 15q14.[1] It is a matricellular glycoprotein and can be secreted by many cells.[2][3][4] Recombinant human thrombospondin is glycosylated with N-linked sugars and produced using baculovirus vectors in insect cells. Human thrombospondin is purified by proprietary chromatographic techniques.
Application
In rat C6 astroglioma cells, TSP1 (thrombospondin 1)-human has been used in coating the cell culture plates to study the effects of integrins on CNTF (ciliary neurotrophic factor) expression.[5] It has also been used in the preparation of secreted factor cocktail for analysis of the human mesenchymal stem cell secretome.[6]
Biochem/physiol Actions
TSP1 (thrombospondin 1) is believed to play a role in cell migration and proliferation, during embryogenesis and wound repair.[7][8] TSP1 expression is highly regulated by different hormones and cytokines and is developmentally controlled. TSP1 stimulates the growth of vascular smooth muscle cells and human foreskin fibroblasts. A combination of IFN-γ (interferon γ) and TNF-α (tumor necrosis factor α) inhibits TSP1 production in these cells.[9] In endothelial cells, it controls adhesion, migration as well as proliferation. It also exhibits antiangiogenic properties and regulates immune processes.[2][3] TSP1 binds to various cell surface receptors, such as integrins and integrin-associated protein CD47.[7] It also plays a crucial role in inflammatory processes and post-inflammatory tissue dynamics.[4] TSP1 has been used as a potential regulator of tumor growth and metastasis.[3] It is upregulated in rheumatoid synovial tissues and might be associated with rheumatoid arthritis.[10] Variants of this gene might be linked with increased risk of autism.[11]
