General description
Fibronectin is a glycoprotein with multiple domains. It comprises a N-terminal, central binding domain and a heparin-binding domain.[1] Bovine plasma fibronectin has polypeptide chains with disulfide linkage and corresponds to a molecular weight of 220 and 215 kDa.[2]
Fibronectins are adhesive glycoproteins and are found both as cell surface proteins and in plasma.[3] Fibronectin (FN) is a multifunctional glycoprotein, the gene of which is localized to human chromosome 2q34-36. This gene spans 75kb and contains 50 exons. It exists as two major isoforms, one soluble and one insoluble form. The former is present in plasma, whereas the insoluble form resides in tissues and extracellular matrix (ECM) of cartilage. There are around 20 different forms of FN due to alternative splicing of a variable region and two type III exons called Extra Domains A and B.[4]
Application
Fibronectin from bovine plasma is used for the following applications:
Used for coating the glass coverslips for the culture of decidual stromal cells (DEC)[5]
Cell movement assays (culture plates were coated with fibronectin)[6]
Cell spreading and immunofluorescence[7]
Protein specificity[8]
Integration of sensor films into microfluidic chips[9]
Histological and Immunohistochemical (IHC) analyses[10]
Used in primary cell cultures from patients with lung carcinoma[11]
Biochem/physiol Actions
Fibronectin is a regulator of protein scaffolding. It coordinates cellular processes like platelet functions, wound-healing, adhesion, migration and aggregation.[1]Study shows that human dental pulp (HDP) cells rely on fibronectin (FN) for initial attachment and subsequent spreading. FN may exhibit mitogenic activity, promoting cell proliferation, and also play a role in regulating the differentiation process of pulp cells into odontoblasts. The presence of FN appears to influence the fate and development of HDP cells, specifically in their differentiation into odontoblasts, which are responsible for dentin formation in teeth.[12]
