Poly-L-lysine hydrobromide has been used to coat:
2-well plates before seeding muscle cells in a research study on rainbow trout myotube formation[1]
flat bottom 96-well plates for in vitro neuronal stem cell differentiation[2]
T75 flasks for primary astrocytic cultures[3]
Biochem/physiol Actions
Poly-L-lysine serves as a coating substance to ensure better attachment of cells to culture surfaces.[4] Research shows that plates coated with poly-L-lysine (PLL) demonstrated an accelerated cell growth rate.[5] Culturing mesenchymal stem cells (MSCs) on PLL-coated substrates has been found to safeguard the stemness properties of these cells and inhibit senescence. PLL-coated plates have demonstrated enhanced MSC growth and up-regulated the expression of genes associated with cell adhesion, differentiation, proliferation, and signaling. Moreover, porous PLL-coated surfaces have shown improvements in cell proliferation and the osteogenic differentiation of MSCs.[5]

Poly-L-lysine is a positively charged amino acid polymer with approximately one HBr per lysine residue. The hydrobromide allows the poly-L-lysine to be in a crystalline form soluble in water. A small amount of product may be found in the beta structure because the HBr interferes with hydrogen bonding between amino and either the carboxyl groups or N or O containing moieties.
